Why GLP-1 drugs cause anhedonia

GLP-1 receptor agonists (semaglutide, tirzepatide, retatrutide) work in part by reducing dopaminergic activity in the reward system. This is exactly why they reduce food cravings and compulsive eating — but it also reduces motivation, joy, and emotional responsiveness across the board. A 2024 meta-analysis found anhedonia symptoms in 23–41% of long-term GLP-1 users.

Key Ingredients

Affron® Saffron Extract — 28mg

The only saffron extract with peer-reviewed RCT data specifically on anhedonia. A 2025 randomized controlled trial showed statistically significant reductions in SHAPS (Snaith–Hamilton Pleasure Scale) scores after 4 weeks of supplementation at 28mg/day. Affron® is standardized to 3.5% lepticrosalides — the active saffron constituents responsible for serotonin reuptake modulation.

Mucuna Pruriens — 300mg (standardized to 15% L-DOPA)

Rhodiola Rosea — 300mg (standardized to 3% rosavins, 1% salidroside)

Adaptogen with a dual mechanism: reduces cortisol (which depletes dopamine) while supporting monoamine oxidase inhibition. Multiple RCTs show improved emotional resilience and reduced fatigue at this dose range.

KSM-66® Ashwagandha — 600mg

The most studied ashwagandha extract on the market. Clinically shown to reduce cortisol by 27.9% and increase testosterone in men — both relevant to the hormonal disruption that often accompanies caloric restriction and GLP-1 use.

Lion's Mane Extract — 500mg (standardized to 30% polysaccharides)

Promotes nerve growth factor (NGF) synthesis, supporting neurogenesis and synaptic plasticity. Emerging research shows positive effects on mood and cognitive function in populations with stress-related cognitive decline.
